When shopping the market for anti-aging supplements it’s practically impossible to avoid the mention of “NAD” and  “NAD+”. But what are these mysterious substances? What is the difference between the two? And, more importantly, what is their relevance to the human aging process? Time to clear the air!

Nicotinamide Adenine Dinucleotide (NAD) is best defined as a cofactor. “Cofactor” is a rather general term indicating any non-protein chemical compound which plays an essential role in catalyzing enzyme reactions. One of the first and most important things to know about this particular cofactor is that it can be found in each and every living cell – not just in humans, but in every single creature on Earth!

Now, onto the issue of that confusing “+” symbol. NAD+ is the oxidized form of NAD which accepts electrons from other molecules and becomes reduced into NADH. This reduced form of NAD is then used to donate electrons.

That’s all a very complicated way of saying that NAD is essential for electron transfer reactions. To give an idea of how important electron transfer is to all life on Earth, it’s worth stating that biochemical processes like respiration, photosynthesis, and oxygen binding are all dependent upon said transfers. In a nutshell, no NAD = no life (as we know it).

This also indicates that NAD+ may be, of all NAD forms, the most direct instigator of electron transfer reactions. For this reason, it is a focal point for the purpose of anti-aging supplementation.

NAD+ and Anti-Aging

Recently, a great deal of research has surfaced which indicates that declining NAD+ levels are a fundamental aspect of the physical and mental decline which is associated with aging.

Among this research is a 2018 study from the Washington University School of Medicine which reviews the current clinical literature on NAD+ and aging. Upwards of 80 studies were analyzed in this review and the researchers conclusion was that “systemic NAD+ decline is one of the fundamental molecular events that regulate the process of aging and possibly limit organismal life span.”1

One study conducted on aged mice concluded that “NMN supplementation exerts significant cerebromicrovascular protective effects in aged mice.” It also goes on to state that clinical trials on elderly human subjects were a necessary and imminent step to take in this line of research.3
